The LABType test applies Luminex technology to the reverse SSO DNA typing method. The target DNA is amplified by PCR using a group-specific primer. The resulting PCR product is biotinylated, allowing its detection using streptavidin conjugated to R-phycoerythrin (SAPE). The PCR product is denatured and rehybridized to complementary DNA probes conjugated to fluorescent microspheres. A flow analyzer, the LABScan 100 (Luminex 100/200) or the LABScan3D™ (Luminex FLEXMAP 3D), identifies the fluorescent intensity of PE (phycoerythrin) on each microsphere. HLA typing assignment is based on the reaction pattern compared to patterns associated with published HLA gene sequences.
